In practice when reservoir parameters such as porosities and permeabilities are estimated by matching reservoir model calculated values to field data, one has some prior information about the parameter values. In addiction, the parameter values are known to be within certain bounds for a particular area. All this information can be incorporated in the estimation method of the simulator by introducing prior parameter distributions and by imposing constraints on the parameters (Tan and Kalogerakis, 1993). These suggested factors for standardization of the Caco-2 model might also be reasons for the variability in published permeability values obtained from different laboratories [86]. In order to avoid a lack of consistent literature-based data, the Caco-2 permeability values of our selected compounds were transformed according to the following scheme the majority of compounds with Papp < 4 x 10 6 cm s 1 were classified as poorly absorbed and assigned a score of —1 compounds with Papp > 8 x 10 6 cm s 1 were classified as well-absorbed and assigned a score of +1. Wohnsland and Faller ([175] performed measurements using a thin (9-10 //in) supported, phospholipid-free hexadecane layer. To validate their model, they used 32 well-characterized chemically diverse compounds. The permeability values obtained with their model could be correlated with known human absorption values if the maximum permeability obtained at different pH was taken into account. However, several disadvantages are related to this method. For hydrophilic drugs, hexadecane by itself has an increased barrier function in comparison with membranes. In addition, the hexadecane layers are not very stable, which makes this assay difficult to apply as a routine screening method. The advantage of this PAMPA setup is that it appears to be a satisfactory substitute for obtaining alkane-water partition coefficients, which are usually very difficult to measure directly, due to the poor solubility of drug molecules in alkanes.
