Methods of Collecting a Pap Smear

Stains used:
– Harris Hematoxylin solution – nuclear stain; has
affinity to the nucleus
– Orange Green 6 (OG 6) – cytoplasmic stain; used
to stain the cytoplasm of mature superficial cells)
– Eosin Azure (EA) – used to stain the cytoplasm of
immature cells (intermediates and parabasals).
The stain contains a mixture of Eosin Y, Bismarck
Brown and Light Green solution

of Light Green solution in each
preparation
• 1. EA 36
• 2. EA 50
• 3. EA 65
Procedure:
• 1. Fixation of smear in 95% ethanol.
• Wash with water
• 2. Primary staining with Hematoxylin
• Wash with water
• 3. Acid/alcohol for (regressive staining); skip this step
in progressive staining
• 4. Blueing with alkalinizing solution
• 5. 1st counterstain using OG 6
• Wash in 95% ethanol
• 6. 2nd counterstain using EA
• 7. Dehydrate, clear and mount.
Precautions in Papanicolaou Staining:
• 1. Leave the smear in solution (alcohol and
water) until the liquid runs off the slides in a
smooth clean sheet.

• 2. Hematoxylin should be filtered daily before


use. Smears should not be allowed to dry in
between any of the staining steps.
