INOCULATION OF MICROORGANISMS FROM THE BODY

INTRODUCTION
Animal and human diseases may be classified, according to the causative agent, as bacterial
diseases, fungal diseases, viral diseases, parasitic diseases, hereditary diseases, and diseases caused by
environmental factors. Frequently, diseases may be brought on by a multitude of causes. A relatively
mild viral infection, for example, if favored by hereditary susceptibility, may then weaken the body’s
resistance to bacterial invasion.
In this activity, you will culture bacteria from different sources in your body. Remember that
bacteria are ubiquitous, so, it is expected that you will also collect them from vital parts of the human
body.

OBJECTIVES
1. To culture bacteria from different sites of inoculation.
2. To be able to detect the presence of microorganisms in the human body.
3. To demonstrate the aseptic technique in the study of bacteria.

MATERIALS AND EQUIPMENT


Prepared Agar Plates Used paper
Sterile cotton swab Tissue Paper
Incubator

PROCEDURE
1. Take out from the refrigerator the culture media that you have prepared in the previous
activity.
2. Remove the excess water that may have accumulated on the surface of the agar media and wipe
dry the Petri dishes. In doing this procedure, try not to open the Petri dishes so as not to expose
the media (opening of the Petri dishes should be done only at the designated area of exposure).
3. Label the Petri dishes that you shall use for inoculation as well as the Petri dishes to be used for
exposure.
4. Your instructor will assign from which particular sites of inoculation and areas of exposure are
you going to obtain your inoculum.

Inoculation
1. The following parts of the body are fertile sources of inoculum.

2. Moisten the cotton swab and smudge on the desired part of the body where you shall obtain
your inoculum (or as assigned by your instructor). Streak smudged the swab onto the surface of
the agar medium.
3. Do not forget to label your streak lines with the source of the inoculum.

4. Wrap all the inoculated Petri dishes with used paper and incubate at 37OC to be used for the
next activity.
