INTRODUCTION
 Known as anti-egg white injury factor.
 Formerly known as vitamin H and vitamin
B7
 found that rats fed large amounts of egg-
white as their only protein source exhibited
neurological dysfunction, hair loss,
dermatitis, and eventually, death.
SOURCES
METABOLISM
 Biotin is found as biocytin in many foods, readily
absorbed.
 Taken up by liver and muscle.
 Bacterial flora will provide adequate quantities
of biotin
 Because it is also synthesised by intestinal flora,
excretion is 3-6 times greater than intake.
ACTIVE FORM
 BIOCYTIN
 Valeric acid chain binds with epsilon amino group
of lysine in enzyme
CO-ENZYME ACTIVITY
 Essential for carboxylation reactions
 ABC of carboxylation reactions
 A – ATP
 B - Biotin
 C – CO2 and Carboxylase enzyme
ACETYL COA CARBOXYLASE
PYRUVATE CARBOXYLASE

• It provides OAA for TCA cycle and

Important in the glucogenic pathway.
PROPIONYL COA CARBOXYLASE
BIOTIN INDEPENDENT CARBOXYLATION
REACTIONS
 Carbamoyl phosphate synthesis in Urea Cycle.
CPS 1 AND CPS 2
 C6 in Purine ring.
 Malic enzyme converting pyruvate to malate.
BIOTIN ANTAGONISTS
 Avidin
 A protein present in egg white- has a great affinity to biotin
 Intake of raw boiled egg causes biotin deficiency
 One molecule of avidin binds with 4 molecules of biotin
 Boiling of egg will neutralize its inhibitory activity
 Streptavidin
USES
ELISA TEST
Biotin
Labelling of
DNA
DEFICIENCY
 During prolonged use of antibacterial drugs
 Symptoms- dermatitis
atrophic glossitis
hyperesthesia
muscle pain
anorexia
split nails
REQUIREMENTS
 30-40µg for adults
 100-300µg in treating deficiency
QUESTION
 Biotin acts as a coenzyme for all except:
a. Pyruvate to Oxaloacetate
b. Acetyl CoA to Malonyl CoA
c. Propionyl CoA to Methyl Malonyl CoA
d. Glutamate to Gamma Carboxy glutamate
