This study evaluates the effects of two decellularization protocols, enzyme-detergent (ED) and detergent-detergent (DD), on the structural and biomechanical properties of human urethral tissue. Urethral samples from 18 individuals were divided into ED (n = 7) and DD (n = 11) groups, with native samples (n = 3) serving as controls. Histological and ultrastructural analyses confirmed that both protocols effectively removed cellular content while preserving essential extracellular matrix (ECM) elements, such as collagen and elastic fibers. Immunohistochemical staining for collagen IV and fibronectin revealed no significant differences between decellularized and native tissues, indicating intact ECM structure. Biomechanical testing demonstrated that DD-treated tissues had significantly lower Cauchy stress (1494.8 ± 518.4 kPa) when compared to native tissues (2439.7 ± 578.7 kPa, p = 0.013), while ED-treated tissues were similar to both groups. Both decellularized groups exhibited reduced stretch at failure and elastic modulus compared to native tissues. Cytotoxicity assays using adipose-derived stem cells demonstrated no signs of toxicity in either protocol. Overall, both ED and DD protocols effectively preserved the urethral ECM structure and mechanical properties, making them suitable for potential use in tissue-engineered grafts and for biobanking purposes. Further research is needed to refine and optimize decellularization methods to improve scaffold recellularization and ensure clinical safety and efficacy. Full article

This study evaluated the use of GFDM transmission in passive optical networks (PONs) by comparing the performance of coherent and non-coherent optical receivers using OptSim 2023.12sp2 and Matlab 2018b ®. The study concentrated on transmitting 10 Gb/s radio frequency signals over optical fiber, emphasizing the significance of high-speed fronthaul links for 5G networks. The findings demonstrated that coherent detection markedly enhances receiver sensitivity by approximately 3 dB compared to direct detection, thereby augmenting the capacity of optical fronthaul networks despite the elevated cost. Additionally, the study recommended investigating pre- and post-compensation techniques to mitigate signal dispersion in optical fibers for further performance optimization. Full article

Background: Monoallelic damaging variants in PHIP (MIM*612870), encoding the Pleckstrin Homology Domain Interacting Protein, have been associated with a novel neurodevelopmental disorder, also termed Chung–Jansen syndrome (CHUJANS, MIM#617991). Most of the described individuals show developmental delay (DD)/intellectual disability (ID), obesity/overweight, and variable congenital anomalies, so the condition can be considered as an ID–overweight syndrome. Case Description: We evaluated a child presenting with DD/ID and a craniofacial phenotype reminiscent of a Pitt–Hopkins syndrome (PTHS)-like condition. We performed a clinical exome analysis on his biological sample, as well as an in silico prediction of the obtained data. At the same time, we interrogated the DeepGestalt technology powered by Face2Gene (F2G), using a frontal image of the proband, and clinically reviewed the earlier CHUJANS patients. In this child, we found a novel PHIP pathogenetic variant, which we corroborated through a protein modeling approach. The F2G platform supported the initial clinical hypothesis of a PTHS-like condition, while the clinical review highlighted the lack of the main frequent CHUJANS clinical features in this child. Conclusions: The unusual clinical presentation of this novel patient resembles a PTHS-like condition. However, a novel variant in PHIP has been unexpectedly detected, expanding the phenotypic spectrum of CHUJANS. Notably, PTHS (MIM#610954), which is a different ID syndrome caused by heterozygous variants in TCF4 (MIM*610954), is not classically considered in the differential diagnosis of CHUJANS nor has been cited in the previous studies. This could support other complex diagnoses and invite further patients’ descriptions. Full article

(1) Background: donor-derived circulating free DNA (dd-cfDNA), an innovative biomarker with great potential for the early identification and prevention of graft damage. (2) Methods: Samples were collected prospectively and the study was performed retrospectively to analyze dd-cfDNA plasma levels in 30 kidney transplant patients during their post-transplant follow-up (15 days, 3, 6, and 9 months), to determine if the result could be of interest in the identification of possible adverse events, especially rejection. The aim was to verify whether the data on sensitivity, specificity, NPV, and PPV compare with reference values and creatinine values. (3) Results: We observed levels of dd cfDNA > 1% in six of nine patients with active rejection (ABMR or TCMR) and elevated values (>0.5%) in two other patients in this rejection group. Our results show low values of sensitivity = 50%, specificity = 61.11%, rejection NPV = 64.71%, and rejection PPV = 46.13% of the technique compared to reference values previously published. With respect to creatinine, only for TCRM, we observed better results for dd-cfDNA in these parameters than in creatinine. Also, our data suggest that dd-cfDNA could help to differentiate those patients with dnDSAs that are going to through rejection better than creatinine, specially at 15 d post transplant. In this study, this appears to have no positive predictive value for borderline rejection (BR) or TCMR IA. (4) Conclusions: plasma levels of dd-cfDNA could be considered an additional or alternative biomarker for graft rejection monitoring in early post-kidney transplant up to several months before its clinical presentation, especially for patients with suspected TCMR or ABMR. We developed an electro-mechanical actuator for use as a steering device for railway vehicles. The specifications for a 50 kN-class electro-mechanical actuator capable of steering control for a sharp of a railway radius curve of up to 250 m, and with Direct Drive motor (DD motor) specifications, were derived as a power source to drive it. The DD motor and an electric mechanical steering actuator equipped with it were designed and a prototype was manufactured. As a result of testing laboratory tests on motor and actuator prototypes, all performance requirements were satisfied. We conducted a field test on the actuator prototype by installing it on a railway vehicle to verify the steering angle performance. The steering action of the actuator worked well when running in curved sections, and the steering angles measured for each curve were in line with the target steering angles. The mean error between the target steering angle and the measured steering angle was only 3.4%, indicating that the wheel steering action of the developed steering system was working very well. Therefore, the developed electro-mechanical actuator is expected to be fully utilized as a steering device for railway vehicles by meeting all the performance requirements of the steering system. Escherichia coli O157:H7, a Shiga-toxin-producing E. coli (STEC), is an important pathogen related to foodborne disease that is responsible for a growing number of outbreaks worldwide and has been detected in processed meats, dairy, and fresh vegetables. Although culturing is the gold standard method for detection of this bacterium, molecular methods based on nucleic acid amplification techniques such as PCR are becoming more common because of their rapidity, sensitivity, and specificity. However, to ensure reliable results among the several alternative PCR protocols (e.g., commercial kits and reference methods), different measurement assurance tools, including validated methods, reference materials, and proficiency tests, among others, are required. Herein, we present a digital PCR method validation for E. coli O157:H7 detection and quantification using seven specific gene sequences; this method quantified nucleic acids from different E. coli serotypes, with a detection range of 6.6 to 7900 copies/µL and a repeatability standard deviation over the concentration range of 1% to 13.6%. The relative standard uncertainty was 3.5–14.6%, and the detection limit was 0.27 copies/µL. Subsequently, two batches of a candidate reference material based on E. coli O157:H7 genomic DNA were then produced and characterized for evaluation of copy number concentration with the validated ddPCR method, with assigned values of 164,770 ± 9251 and 172 ± 9 copies/μL. Thus, this study demonstrated the development of a validated method and reference material for dPCR and qPCR detection of E. coli O157:H7, a key STEC responsible for food poisoning. Full article

R-RDSP: Reliable and Rapidly Deployable Wireless Ad Hoc System for Post-Disaster Management over DDS

After natural disasters such as earthquakes, floods, or wars occur, cellular communication networks often sustain significant damage or become impaired. In these critical situations, first responders must coordinate with other rescue teams to communicate essential information to central command and survivors. To address this challenge, we have developed a reliable and rapidly deployable wireless ad hoc system for post-disaster management using Data Distribution Service (DDS) middleware, specifically RTI-DDS, named R-RDSP. The R-RDSP further enhances these metrics, achieving a 14.5% improvement in end-to-end delay and a 20.24% improvement in round-trip delay over the RDSP scheme. The R-RDSP system consists of three main modules: client, relay, and server. Each module connects to others via an ad hoc network, ensuring direct device-to-device communication without relying on existing infrastructure. The client module collects and sends the victim’s location and emergency messages. The relay modules forward these messages across the ad hoc networks, ensuring minimal delay and high reliability. Finally, the server module receives the messages, processes them, and coordinates the response. Leveraging RTI-DDS for reliable message distribution, the system demonstrates robust performance even under challenging network conditions. Full article

Digital dermatitis (DD) poses a major animal welfare concern for the dairy industry, with even broader economic implications for the agricultural industry worldwide. The postbiotic, a Saccharomyces cerevisiae fermentation product (SCFP), has had a positive influence on the innate immune system of cattle, which makes it a potential candidate as a feed supplement as part of a prevention strategy for DD. This study investigated the effect of a commercial SCFP feed supplement compared to a control feed supplement on the production of pro-inflammatory cytokines (IL-1β and IL-6) by peripheral blood mononuclear cells (PBMCs) in Holstein Friesian steers experimentally infected with DD. The results showed that SCFP supplementation was associated with an overall reduced IL-1β production (p = 0.005), particularly prior to experimental inoculation with a DD lesion homogenate. However, the results of the analysis suggest that the innate immune system in the SCFP group became prepared to respond more rapidly to DD infection post-inoculation. During active (M2), chronic (M4), and focal flare-ups (M4.1) of DD, SCFP supplementation resulted in a more rapid secretion of IL-1β (M2: p = 0.038; M4/M4/1: p = 0.034). A more rapid response to DD infection for IL-6 was only found for chronic (M4) and focal flare-ups (M4.1) of DD (p = 0.006). These findings emphasize the difference in cytokine response between various stages of DD in the SCFP group compared to the control, highlighting implications for DD prevention and treatment. The outlet flow rates and changes in behaviors of five outlet ports where water and air–water (two-phase) mixtures pass horizontally in a manifold pipe system were investigated experimentally. The effects of different air-flow rates, vacuumed from the atmosphere with a Venturi device in the system, on the outlet flow rates and diameters of the manifold port outlets were compared by measuring the outlet jet lengths. The system performance provided homogeneity of manifold port outlet flows and was tested. As a result, it was observed that homogeneous jet lengths were obtained in both single and two-phase low main manifold flows and equal outlet port diameters. When the main manifold flow rate V is 1.5–2 m/s, the system is stable and produces high jet lengths. The manifold pipe systems used in the experimental setup provide suitable working conditions for d/D = 0.433. The system does not show a smooth flow pattern with Venturi devices for d/D < 0.433. The low flow rates in this study’s tests are key. They are vital for designing micro irrigation systems. This depends on the critical d/D ratio of the system. Full article

Short-Term Impact of Digital Mental Health Interventions on Psychological Well-Being and Blood Sugar Control in Type 2 Diabetes Patients in Riyadh

Background: Type 2 diabetes (T2D) management is complicated by psychological factors, yet mental health interventions are not routinely integrated into diabetes care. This study investigated the impact of a digital mental health intervention on psychological well-being and glycemic control in T2D patients. Methods: A quasi-experimental study was conducted with 120 T2D patients divided into intervention (n = 60) and control (n = 60) groups. The intervention group received a one-month digital mental health intervention alongside standard care. Psychological well-being (PHQ-9, GAD-7, and DDS) and glycemic control (HbA1c) were assessed at baseline and post-intervention. Results: The intervention group showed significant improvements in HbA1c levels (−0.5%, p = 0.032), PHQ-9 (−3.1, p = 0.001), GAD-7 (−2.8, p = 0.006), and DDS (−7.7, p = 0.012) scores compared to the control group. Strong correlations were observed between psychological improvements and HbA1c reductions. Higher engagement with the digital platform was associated with greater improvements in both psychological and glycemic outcomes. Conclusions: Integrating digital mental health interventions into T2D care can significantly improve both psychological well-being and glycemic control. These findings support a more holistic approach to diabetes management that addresses both mental and physical health aspects. Full article

Background/Objectives: Interest in drug delivery systems (DDS) based on inorganic substrates has increased in parallel with the increase in the number of poorly water-soluble drugs. Hydroxyapatite is one of the ideal matrices for DDS due to its biocompatibility, low cost, and ease of preparation. Methods: We propose two doped hydroxyapatites, one with Ba on Ca sites another with Si on P sites, with the aim of improving the dissolution rate of piretanide, a diuretic, poorly water-soluble drug. The hybrids were characterized by different physical–chemical techniques, and their formation was demonstrated by infrared spectroscopy, thermal analysis, and electron microanalysis, as well as by comparing the results with those obtained on physical mixtures of HAPs and properly prepared piretanide. Results: Both the hybrids improved the piretanide dissolution rate compared with the physical mixtures and the drug alone. The dose was completely solubilized from the Si-doped hybrid in about 5 min in the three fluids considered. This remarkable improvement can be explained by an increase in the wettability and solubility of the drug loaded in the drug-carrier systems. Conclusions: Different experimental techniques, in particular spectroscopy and electronic microanalysis, proved the successful loading of piretanide onto doped HAP. Pharmaceutical measurements demonstrated rapid drug release in different fluids simulating gastrointestinal conditions after oral administration. These hybrid systems could be a very promising platform for drug delivery. Full article

Haematococcus pluvialis contains valuable bioactive compounds, including astaxanthin, proteins, and fatty acids. Astaxanthin is known for its various health benefits, such as preserving the redox balance and reducing inflammation. However, its low stability and poor water solubility present challenges for various applications. Hot-melt extrusion (HME) technology enhances the aqueous solubility of H. pluvialis extracts, increasing the usable astaxanthin content through nanoencapsulation (HME-DDS-applied extracts, ASX-60F and ASX-100F). This study compared the effects of HME-DDS-derived extracts (ASX-60F and ASX-100F) and the non-applied extract (ASX-C) under inflammatory and oxidative stress conditions. In animal models of sepsis, 60F and 100F treatment exhibited higher survival rates and a lower expression of pro-inflammatory biomarkers compared to those treated with C. In lipopolysaccharide-stimulated RAW 264.7 macrophages, nitric oxide (NO) production and the expression of pro-inflammatory mediators such as cyclooxygenase-2 and inducible NO synthase were reduced by 60F or 100F treatments via ERK/p-38 mitogen-activated protein kinase (MAPK) signaling. Moreover, 60F or 100F inhibited reactive oxygen species production regulated by nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) signaling. Collectively, these findings suggest that HME-DDS-derived H. pluvialis extracts exert anti-inflammatory and antioxidant effects by inhibiting MAPK phosphorylation and activating Nrf2/HO-1 expression. Plasmopara viticola is the causal agent of Grapevine Downy Mildew (GDM), which is a devastating disease of grapevines in humid temperate regions. The most employed method for protecting grapevines against GDM is the application of chemical fungicides. In Spain, Carboxylic Acid Amides (CAAs) are a fungicide group currently utilized in GDM control. In P. viticola, resistance to CAAs is conferred by G1105S and G1105V mutations in the CesA3 gene. Droplet digital polymerase chain reaction (ddPCR) is an innovative technique that combines PCR and droplet microfluidics to disperse the sample into thousands of water-in-oil droplets in which an amplification reaction is individually performed. In this study, we set up a ddPCR protocol to quantify S1105 and V1105 mutations conferring resistance to CAAs in P. viticola. The optimal PCR conditions were established, and the sensitivity and precision of the protocol were assessed. Four P. viticola populations coming from commercial vineyards in northern Spain were analyzed, and different allele frequencies were found in the analyzed samples corresponding to the different fungicide management strategies, ranging from 7.72% to 100%. Knowing the level of mutated alleles allows for designing resistance management strategies suited for each location. This suggests that similar ddPCR assays could be developed for studying mutations implicated in fungicide resistance in other fungicide groups and plant pathogens. Full article

This paper presents an advanced modular modeling approach for vertical vibration analysis of dynamic systems using the Generalized Receptance Coupling and Frequency-Based Substructuring (GRCFBS) method. The focus is on a four-DoF half-vehicle model comprising three key subsystems: front suspension, rear suspension, and the vehicle’s trimmed body. The proposed technique is designed to predict dynamic responses in reconfigurable systems across various applications, including automotive, robotics, mechanical machinery, and aerospace structures. By coupling the receptance matrices (FRFs) of individual vehicle modules, the overall system receptance matrix is efficiently derived in a disassembled configuration. Two generalized coupling methods, originally developed by Jetmundsen and D.D. Klerk, are employed to determine the complete vehicle’s receptance matrix from its subsystems. Validation is achieved by comparing the results with established methods, such as direct solution and modal analysis, demonstrating high accuracy and reliability for complex dynamic systems. This modular approach allows for the creation of reduced-order models focused on key measurement points without the need for detailed system representation. The method offers significant advantages in early-stage vehicle development, providing critical insights into system vibration behavior. Full article

Efficient early pathogen detection, before symptom apparition, is crucial for optimizing disease management. In barley, the fungal pathogen Pyrenophora teres is the causative agent of net blotch disease, which exists in two forms: P. teres f. sp. teres (Ptt), causing net-form of net blotch (NTNB), and P. teres f. sp. maculata (Ptm), responsible for spot-form of net blotch (STNB). In this study, we developed primers and a TaqMan probe to detect both Ptt and Ptm. A comprehensive k-mer based analysis was performed across a collection of P. teres genomes to identify the conserved regions that had potential as universal genetic markers. These regions were then analyzed for their prevalence and copy number across diverse Moroccan P. teres strains, using both a k-mer analysis for sequence identification and a phylogenetic assessment to establish genetic relatedness. The designed primer-probe set was successfully validated through qPCR, and early disease detection, prior to symptom development, was achieved using ddPCR. The k-mer analysis performed across the available P. teres genomes suggests the potential for these sequences to serve as universal markers for P. teres, transcending environmental variations.
