Harvesting Multi-cellular Microtissues from the 3D Petri Dish®

1. Remove cell culture medium surrounding the 3D Petri Dish® containing the multi-cellular

microtissues.

2. Place fresh cell culture medium into the well of a new 12 or 24 well plate depending on the

size of the 3D Petri Dish® used (1.0mL for 12 series or 0.5ml for 24 series).

3. Carefully remove the 3D Petri Dish® with a sterile spatula or sterile tweezers. Invert the 3D

Petri Dish® into the well of the new plates. Be careful to not create air bubbles between the

upside down 3D Petri Dish® and the bottom of the new culture plate. Cover the plate and

place the 6-well plate in a swinging bucket rotor in a centrifuge and spin for 5 minutes at 500

rpm.

4. After centrifugation, view the inverted 3D Petri Dish® under the microscope to ensure that

the multi-cellular microtissues have been released.

5. Remove the empty 3D Petri Dish® with a sterile spatula or sterile tweezers.

6. The harvested multi-cellular microtissues remaining in the cell culture medium can be

washed and further processed depending on the application. (Immuno-staining, Western blot,

RT-PCR, etc).
