1.0 PURPOSE
1.1 To lay down the procedure for operation and calibration of Equiptronics EQ-
621 microcontroller base pH meter.

2.0 SCOPE
2.1 This procedure shall apply to Equiptronics EQ- 621 microcontroller base pH
meter.

3.0 RESPONSIBILITY
3.1 Staff member/ Lab Incharge are responsible to perform the activity as per
procedure.
3.2 HOD are responsible for implementation of the procedure.

4.0 REFERENCES
4.1 SOP is prepared with referenced to operation manual of Equiptronics EQ-
621 microcontroller base pH meter.

6.1 How to use microcontroller base pH meter model EQ- 621


6.1.1 Put on the switch provided at the back of instrument.
6.1.2 First the instrument well display brand , model and version after that it
well check whether ATC probe connected or not
6.1.3 If ATC probe is found the instrument will go in ATC mode without any
warning else the instrument will display the message “ATC mode not
present “, “transferring to MTC mode “& after that temp. setting menu
will display with pre- settedtemperature at 25 degree cel.
6.1.4 This set temperature can be changed ( temperature of liquid ) using
up& down keys &then press ´measure / recall” key to read pH value
6.1.5 The instrument now display real pH value & sated temperature
showing “MTC” at the end of second line
6.1.6 In the main menu, up& down keys will disabled.
6.1.7 Only measure / recall & store keys from in action.
6.1.8 On placing measure / recall the instrument goes in recall PH mode

6.1.9 The earlier store value of pH can be their location
6.1.10 To comeback in the main menu , press measure / recall key display
will now show actual PH
6.1.11 On pressing of “store “ key , the instrument will display of a message
“are you sure you want to store . “
6.1.12 The reading will store on pressing the “store” key again
simultaneously showing location
6.1.13 Instrument will take care of location automatically & increment up to
9& then come to 00 locations.
6.1.14 If you do not want to store , press “measure /recall” Key to come back
to the main menu
6.1.15 In case you press “calibrate “in any mode the instrument will promote
“please select calibration mode
6.1.16 It will not allow you to change calibration until you select
6.1.17 Hence it will be locked.

6.2 How to check with electronic buffer:


To cross check in case of faulty reading, we have provided electronic buffer .
The instrument is to be use in MTC mode i.e without temp. Probe special
connecting cord is to be used in place of PH electrode bring the instrument in
“actual pH “mode and connect to respective pH buffer sockets. The
instrument will display respective PH this means that instrument is OK, and
fault is with electrode.
Calibration can also be done with electronics buffer where ± 5 PH
accuracy tolerated.
N.B: there can be fluctuation in the reading if the sole. Are not
mixedproperly. the fluctuation will stop as soon as mixture becomes
homogeneous.

Periodic calibrations are to be done as follow:


1. New electrode - daily for one week
2. After 1 weak of usages - twice per week
3. After 4 week of usages - once per 3 months

Standard Operating Procedure


The self-life of unused electrode is six month. The electrode works better
and life may be extended to two to three years if used daily or stored in wet
condition. Hence it is advisable to wrap moist cotton in plastic bag for wet
condition or use wetting cap that is provided with the unbreakable electrodes
Caution: Connect PH probes and temperature probes in their respective
sockets . Interchanges will result in damage to the probes
6.3 Measuring pH in ATC mode: (The instrument has to been pre-calibrate)
6.3.1 Connect pH and temperature in their respective sockets in the
instruments & switch on the power
6.3.2 After getting actual ph. and temperature display intactmode, dip
probe and electrode in the solution and measure.
6.4 Measuring pH in MTC mode (The instrument has to be pre calibrated)
6.4.1 Connect electrode to the instrument and switch ON power. The
instrument will go into MTC mode and display ‘set Temperature at
25*c’ which is default.
6.4.2 Press ‘up down” switch to change the temperature value of the
solution (whose temperature is known ) and press “measure /Recall
”switch to set the temperature.
6.4.3 Dip the electrode in the solution and measure pH.
6.5 Measuring pH in mV mode
6.5.1 The instrument is pre –calibrated in our factory mode.
6.5.2 connect the required probe in the PH socket of the instrument and
switch on the supply .
6.5.3 The instrument will go into PH mode and stop there .the PH and
Temperature values are not significant in mv measurement .
6.5.4 press mode switch to bring the instrument in mV mode .Dip the
electrode in the solution and measure the mV .

Sronging:-When the instrument is in ‘Actual pH Mode’,press ‘store


button .The instrument will ask if you want to store button again.It will
store and display location of storing.

To Recall:-Press “Measure /Recall ’’button .The stored values will be
displayed with locations.

6.6 Preparation of buffer for very accurate measurement


6.6.1 Prepare double distilled water. Keep it closed to avoid Co2 mixing.
Preferable boil it before use.
6.6.2 Take a clean beaker (cleaned bye distilled water) and place buffer
powder (standard brand) or tablet in it. Now add above prepared
distilled water so that the total volume is 100ml. Allow it to dissolve
and cover it at the same time to avoid Co2 mixing. The buffer so
prepared will be very accurate. (as specified by manufacture)
6.7 Some hints for stable readings
Of all the instrument pH-electrode system requires great understanding and
care.
We incorporate from time to time latest information gathered by us on the
above subject. If the fluctuation does not stop then try the following:
6.7.1 Check the level of KCL. Add freshly prepared KCL each time if
possible. The KCL has to follow out of fibre junction freely.
6.7.2 See that the tip of the fibre junction dips in the solution. 3. Remove
any bubble in the glass bulb.
6.7.3 Prick rubber cap for free flow of air, or open the cap and close it for
pressure equalisation.
6.7.4 Clean both pin and socket of electrode with alcohol using small
brush. Blow dry air
6.8 Calibration Procedure:
6.8.1 Ensure that instrument is clean and free from dust.
6.8.2 Switch on the instrument and rear pan to ‘on’ position.
6.8.3 Connect glass and reference electrode to appropriate at the real
panel.
6.8.4 Set the temperature knob to temperature of solution
6.8.5 place electrode in buffer 7 .

6.8.6 Press” stand by / “ read “ switch on the front panel to “ read” .
6.8.7 Adjust ASM ( asymmetric potential knob) control knob to read 7 ph in
the display .

6.8.8 Bring the “ stand by” / “ read “ back to “ stand by “


6.8.9 remove the electrodes from buffer 7. Wash and place them in buffer 4
or buffer 9.2.
6.8.10 Press the “ stand by’’ / “ read “ switch to “ read “.
6.8.11 Adjust SLP Control knob ( stope control knob) to read 4ph or 9.2 ph
in the display .
6.8.12 Check back buffer 7 & adjust ASM control if necessary .
6.8.13 Repeat 7 & 4 or 9.2 buffer till you read both buffer values correctly.
6.8.14 Rinse the electrode with distilled water & place it in sample and
sample ph.
6.8.15 Precaution
6.8.15.1 Do not leave electrode dry, when not in used keep electrode
in distilled water.
6.8.15.2 Do not wipe the glass electrode with coarse material used
only nice filter or tissue paper.
6.8.15.3 The buffer should not contaminate & should not be used
after 3 months.
